There are many ways how to explain that:

1) The first thing the came into my mind when I saw the screenshot from the newsletter was, that it could be catacombs under Mystrock. The skulls doesn't really fit into this, but still...

2) He could simply escape.

3) Maybe, when the change to non-human happened, he was outside of Mystrock(although from what I have understood from MM so far, people from Mystrock rarely leave the city, especially nobles).

3) is very true.
In fact Mystrockians can leave the town but would usually stick around in the "mystrock territory".
Reasons to go through the cave to the South would be exceptionnal.
And the bridge to the East kinda marks the frontier between the ok lands and the crappy peasant lands.

as for 1), who would those big skulls belong to?

Tbh, it didn't occur to me, that these skulls could be genuine. If so, then they would obviously belong to some giant, probably from an ancient, extinct or very rare race (we haven't met any giants in MM so far).
The place would probably be some ancient crypt, then. Maybe Marine is looking for some artifact here?

Or just huge statues. However, there are also a lot of normal sized skulls.

Maybe they are searching for the legendary Udo in a crypt.

Maybe someone will need her help. She already did the same in MM3 when she helped Jack and Salome, despite the fact that time was running out.

however this time there really is VERY little time +::O_o
